Manchester United's Cristiano Ronaldo and Nani will miss Portugal's friendly against Greece on Wednesday.
The in-form pair, who both scored as United beat Liverpool 3-0 on Sunday, are suffering from leg muscle problems.
Henrique Jones, Portugal's team doctor, said: "We proposed to coach (Luiz Felipe Scolari) that the players be released from the squad and he accepted."
There was some good news for Scolari, however.
Ricardo Oliveira and defender Ricardo Carvalho have recovered from injuries and are available for the Euro 2008 warm-up match in Dusseldorf, Germany.
Oliveira had a shoulder injury, which caused him to miss last weekend's game with his Real Betis team in the Spanish league, while Carvalho injured his head during Chelsea's game with Arsenal on Sunday.
Portugal have not called up replacements for Nani and Ronaldo.
