Danilo Di Luca's home was raided by Italian police but the Giro d'Italia runner-up was adamant he had not used banned substances.
The International Cycling Union said Di Luca had been provisionally suspended after testing positive for the banned blood booster CERA during May's Giro.
An investigative source in Rome said searches had taken place at Di Luca's house near Pescara and the home of a doctor accused of supplying athletes with illegal drugs.
The LPR rider was banned for three months in 2007 for his links to the doctor.
Di Luca, the 2007 Giro champion, earlier told Sky television there could have been an unintentional mix-up with his test and he wanted his B sample to be checked at a different laboratory.
"I hope the truth comes out and the truth is that I haven't taken a drug, I haven't taken this drug," he said.

I'm sure when that B sample comes through he will be cleared and this will all have been a terrible mix up, what a t**t, just hold your hands up and admit it, still at least he didnt run away and hide behind a car like Ricco
I like diLuca...hope he's cleared.
Bit of a bummer for DiLuca but I'll bet someone else will be sweating who was pretty close to his wheel on those Giro stages!
Thought he has already retired? Oh yeah, we waiting for the B sample positive. Too late slow boat Italian police... its all long gone flushed down the WC.
I hope he is right!!
They always deny it at first, the same thing was with Ricco. Im not saying he doped but i guess we will have to wait for the outcome.
I believe him...
Everybody down on the floor!!!!
