The Giallorossi will be without their talismanic captain Francesco Totti - who netted twice in Sunday's 2-2 draw with Juventus - as the striker is suffering from a thigh strain.
Roma are also without Italy midfielder Simone Perrotta, who has 'flu, and injured right-back Marco Cassetti, who is expected to be sidelined for at least three weeks and is likely to miss Saturday's clash with champions Internazionale as well as next week's Champions League match at Manchester United. Fit-again Italy defender Christian Panucci is likely to take his place.
Fiorentina's Romania striker Adrian Mutu warned Roma that the Tuscan side will be aiming to knock them off the top spot at the Stadio Artemio Franchi.
"We cannot compete for the title yet, but we can go top, even if it's just for one day," the former Chelsea forward said.
Roma have 10 points from four games, two more than Fiorentina, Inter and Atalanta.
Elsewhere, Brazilian defender Maicon misses Inter's home game against Sampdoria after receiving a three-match ban for kicking Erjon Bogdani during Sunday's 2-2 draw at Livorno.
Centre backs Marco Materazzi and Cristian Chivu are also out with thigh and shoulder injuries respectively, and Colombian defender Nelson Rivas is unavailable after passing out in training at the weekend.
France midfielder Patrick Vieira is still unfit with a hamstring injury.
Juventus will be stretched at the back when they entertain Reggina after Portuguese defender Jorge Andrade broke his kneecap at Roma on Sunday.
In defence, Juve boss Claudio Ranieri is also without Jonathan Zebina, who is suspended, and Jean Alain Boumsong, who has a thigh muscle injury.
Centre-back Sebastiano Siviglia will not play in Lazio's home game against Cagliari, after fracturing three fingers in training on Monday.
The Rome club, who play host to Real Madrid in the Champions League next week, are seeking their first win of season in Serie A.
Striker Alberto Gilardino is expected to take Filippo Inzaghi's place in AC Milan's starting line-up at Palermo. The European champions are aiming to end a run of three consecutive draws with a win in Sicily.
