Cook criticised his players following the weekend defeat to Wycombe, saying they lack fire and hunger before branding them "soft and weak".
Defenders Neal Trotman and Terrell Forbes, winger Jack Waddle, who has joined Sheffield FC on loan, and forwards Scott Boden and Craig Westcarr, who is now back at the Proact Stadium after his loan spell at Walsall ended, have already been placed on the transfer list.
And after a third npower League Two defeat in five matches at Adams Park, more changes are expected in an attempt to keep the Spireites' promotion hopes alive.
"We want to get promotion and, unfortunately, at the minute the ingredients aren't right," said Cook.
"Sometimes at a football club personnel must change. How quick we can change, that depends. We haven't got a magic wand that can bring the right players in.
"I look at the patterns, the results and what we do this season and they say that we concede goals and we're inconsistent.
"Whether it is (former manager) John Sheridan, (assistant manager) Tommy Wright or Paul Cook, the pattern is the same and it's the personnel in the dressing room. I haven't been able to change it, I hoped I would."
