On-loan midfielder Lundstram suffered an ankle injury during the 1-1 home draw with Portsmouth last weekend, with the problem forcing the Everton teenager to withdraw from the England Under-19 squad for Thursday night's friendly against Turkey.
Lundstram returned to Goodison Park for treatment and Rovers boss Brian Flynn is awaiting the verdict.
"We're hoping for good news on John," said Flynn. "He was disappointed to miss out for England but he was quite positive when I spoke to him last Sunday.
"Everton were quite confident he'd be okay from the contact I've had with them and it was best all-round that he went back to them for treatment."
Without Lundstram, Flynn would be short in midfield as another loanee Dean Furman is away on international duty with South Africa and David Syers (ankle) is still injured.
With promotion rivals Sheffield United and Brentford kicking off at 12.45pm on Saturday, Doncaster could find their lead at the top of the table cut down to one point or have been replaced at the summit by the Bees by the time they start at Glanford Park.
