Australian Brookes was pipped to top spot in opening practice by Tommy Hill, but the Swan Yamaha rider was down in sixth in the second run-out.
Alex Lowes on the WFR Honda was second to Brookes, around a quarter of a second off the pace.
Brookes’s team-mate Alastair Seeley was again third.
Hill narrowly leads the championship from Rapid Solicitors Kawasaki rider Shane Byrne, who was fifth in the second session, and Brookes.

Tommy Hill snatched top spot in a wet opening practice for the British Superbike weekend at Oulton Park in the final moments ahead of Josh Brookes.
Australian Brookes, who was the first rider to head out in the delayed session, looked set to finish top of the timesheets before defending champion Hill put the Swan Yamaha there with an effort of a minute 49.347 seconds.
Brookes was 0.213s down, with fellow Tyco Suzuki rider Alastair Seeley third, over half a second off the pace after crashing late on.
Scott Smart (Moto Rapido Ducati) was fourth, with Shane Byrne (Rapid Solicitors Kawasaki) fifth.
Alex Lowes of WFR Honda spent time in top spot but finished over a second behind the eventual leader in sixth.
Jon Kirkham was again unlucky in crashing out on the Samsung Honda at Knickerbrook while Patric Muff’s session was also ended early.
