Toulon centre Contepomi, his team-mate Fernandez Lobbe and Stade Francais prop Roncero have been named by coach Santiago Phelan from a 46-man preliminary squad ahead of the match at the Estadio Monumental Jose Fierro in Tucuman.
Contepomi has returned to the national squad after a one year absence and said: "I'm focused on this tour because I've been absent for a long time and I want to enjoy these moments with the Pumas."
He added: "We have to get the most out of these matches because we will not play too many games (before the World Cup)."
The former Leinster man looked forward to playing Scotland, a team they will face in the World Cup next year.
He said: "It helps you know them better. It is also an advantage for them so we must focus on our play and train a lot in order not to commit mistakes."
Leicester duo Horacio Agulla and Marcos Ayerza, Albi winger Lucas Borges and London Irish scrum-half Alfredo Lalanne are other important inclusions in the squad.
Only four of the squad play in the regional leagues in Argentina - third rower Genaro Fessia, CASI scrum-half Agustin Figuerola, Universitario La Plata second rower Mariano Galarza and San Luis hooker Agustin Creevy.
The starting XV will be revealed on Tuesday evening, with the second Test will be played in Mar del Plata on June 19.
